[ Saturn ] Can I Unplug Controllers Even If It's Not Off?

Hi. My japanese Saturn's port 1 is busted, only port 2 works. Is this going to damage the system: I use my Nights Into Dreams Controller (on plus sign mode) then I switch to the gun.
By the way, when do you use the plus and zero sign in the Nights Into Dreams controller?
